The Neapolitan Mastiff is a giant breed, all giant breeds are plagued by Heart Problems.  Perhaps it is because they have the biggest hearts and give the most love.  Although there exists no definitive data on the Neapolitan Mastiff, it would seem the most common Heart ailment in the breed is Cardiomyopathy.
